This puzzle can be solved in two ways. First, you can play all the notes in a row, which is the easiest way to solve it.

Flex three times, but be care to dodge his attacks as he will get stronger with each flex. He will try to hit you with a lightning bolt on his third flex. If you dodge it, you will be able to avoid the attack.
If you do not dodge the lightning strike, it will deal a lot of damage to you, so be careful. You will have to fight him for a while until he is defeated. When you are done fighting him, go back to the entrance of the cave and go through the door. Go down the stairs and follow the path until you reach a fork. Take the left path and then take the right path.
The path you take will lead you to a room with two doors on either side of it. Enter the first door and open it to find a treasure chest. Open the second door to get a key. Now go up the ladder and enter the next room.
